I need to add bus routes to a map. What are my best options?

Answer:

It depends on what you need to do with these routes. If you need these to be "real routes" in MapPoint (so you can use the distance calculations and other attributes), you need to drop as many pushpins as you need to get MapPoint to calculate the right route. Just adding a start and end probably won't do it, as MapPoint will find the shortest/quickest route which is usually not what a bus route is. The only way around this is to add as many pushpins as it takes to ensure that the actual bus route is the shortest/quickest connection between the dots.

If you're only concerned about having lines on a map as a visual representation of the routes, your best bet may be to use the drawing tools to add colored lines. And if you need multiple routes displayed simultaneously, using the drawing tools is your only choice (you can only have one route displayed on the map at any given time).
